Pharmacokinetics:

Methyl cobalamin:

·         Methyl cobalamin binds with an intrinsic factor and form a complex which is absorbed in distal ileum.

·         Elimination - Bile.

·         Half-life - 6 days.

 

Folic acid:

·         Absorption - Rapid absorption mainly from the duodenum and jejunum.

·         Extensively bound to plasma protein.

·         Principal site of storage - Liver.

 

Pyridoxine:

·         Pyridoxine in the body varies from 16 to 25 mg.

·         Half-life -15 to 20 days.

·         This metabolite is excreted mainly in the urine.

 

MOA:

Methyl cobalamin:

  • Methyl cobalamin plays an important role. Orally administered vitamin B12 binds to intrinsic factor (IF) during its transport through the stomach. The separation of Vitamin B12 and IF occurs in the terminal ileum when calcium is present, and vitamin B12 is then absorbed into the gastrointestinal mucosal cells.           

Folic acid:

  •  Folic acid acts as a cofactor for trans formylation reactions in biosynthesis of purines and thymidylates of nucleic acids. 

Pyridoxine:

  •  Vitamin B6 (Pyridoxine) acts as a coenzyme in the metabolism of protein, carbohydrate, and fat.
